A few of the simpler, nondescript ones, anyway. Mostly I use the ones that aren't city-centric, like the Survivor glass or the one that says "James's" on it; that, or the ones where I have far too many, like ones from Alaska or Mexico (my parents have gone on a lot of cruises).
It was either San Francisco or Baha California in Mexico, but it was a simple, small trinket that I could collect to say I was there. And yes, this was a couple years before I began my drinking career in earnest.
Also, like others have said, they're far more practical than commemorative plates.
Haha, one of my offline friends is currently obsessed with Assassin's Creed 2 as well. Have you used all of those shot glasses at some point or another?
All I'm saying is, I would never stack all those shot glasses on a large, sturdy tray, then try to take it across the street during a police chase. The streetwise bad guys will jump over them, but the squad car in pursuit WILL drive through them.
I don't drink, but I still think shot glasses are cute. I actually have a few that I've picked up over the years. Nothing like your collection though. XD

Niiiiiice! My brother does this, as well as collecting beer coasters from around the world. That's a sweet collection you have there!
