I'll try to answer the best I can.
1) A guest poster is someone who has permission to post in a world that another person has created. For example, if you gave permission to me to post in this world, 'Opinions, Anime and lols:3' then I'd be a guest poster!
2) Don't know about #2, perhaps try using google Chrome?
3) Signatures (as in what you see at the bottom of someone's comment)? If it's that then only members can have them and you can edit what your signature is under profile. As for comments, pressing the 'reply' is correct, but if nothing happens, I suggest google chrome (it has helped a lot of Otakus, including me! :D). Hope that helped!

For example: I can respond to things you post in your world "Opinions, Anime, and Lols:3" by commenting like I'm doing right now. However, if you allow me on as a guest poster I can actually post my own topics here. Lots of people make worlds so they can role play stuff and thus they allow their friends to guest post in their world or there are worlds made where artists can ask advice about lots of different art related topics. "Artist Cafe" is one such world of which I am a member.
2. Erm, I don't really use the chat so I have no idea. Sorry ^^'
3. If someone writes on your wall or comments on a piece of art you've uploaded you'll get an alert in your "backroom." It should be indicated to the right of either "Private Messages" or "New Comments." If you click on either of those it will take you to a page that shows all the comments/messages that you've received. If you click on the message under the "Snippet" heading it'll take you to the message. All you have to do is click "Reply" which should be under the person's name and to the right.
Still, it seems that you're able to get to reply but it won't do so? Are you clicking the "submit comment" button when you're done? Because if you click "close comment" it doesn't upload your comment, but actually closes your comment and lets you look at what you're replying to without actually losing your message. Maybe you weren't clicking the right button? I know I did that for awhile before I got it figured out -^^- If that's not it maybe it's your internet connection?
