As of now, no she isn't his sister. She is a random cousin. Her reason for surviving is also spoilerific, but it is not because Itachi had a soft spot for her or because she wasn't around during the genocide.

This is going to be a response to both of your comments.
Originally Raines hair looked nothing like that, however as I was devloping Raine, to make her hair anotomicly correct her hair ended up like that. Its hard to understand unless I show you how it got that way. but thats a lot of effort im not willing to give you. Raine and Eds hair are a lot a like but there are also differences. Raines tionk is in the front, not that back like Ed's. Raine's is also poofier and sticks out to the sides more to were you never see her ears. Ed's are noticable when i manage to draw it right.

Next: Raine, is not a freaking vampire! Her eye color means NOTHING!! Thats just her eye color. Her 'fang' symbolize her playfulness and trickery. She doesnt drink blood or will die by sunlight.

And also, on the chapter page, Raine looked like a vampire... fang and weird coloured eyes...

It just occured to me that raines hair looks like Edwards from FMA... maybe you really DO have it on the brain...
