nika and embrose's great grandma was a member of the uchiha clan. she broke away and married a member of the darkness village's lead clan. then her daughter(their mother) married into the rosekei clan. nika got the sharigon chakra and it mixed w/ her demon's chakra, so that's why it's opposite sasuke's.
